Written in what?? A 'trumpet' part is written in Bb...If you're using a C-trumpet going to Bb music, you lower it a full step. If you can get access to a keyboard you can figure it out fairly easily.

2. If I have a trumpet part written in E and I use a B-flat trumpet I know that I transpose up a tritone (Augmented 4th) or 6 half steps right. What do you do if it is a C trumpet is it up a major third (3 half steps up)?
Oh, boy. I'll trust the older guys (assuming the people who post like they know what they are talking about are older) to have that one right. Too much for my brain.
It's written in "do" - pronounced "doe" as in "do, a deer" and "do, re, mi". "Do" means it's written in the key of C and should be transposed up a whole tone to be played on Bb trumpet.
